Plexxikon is pursuing a number of drug targets for the treatment of metabolic disease, with lead programs for type 2 diabetes and obesity indications.
PPM-204 (PLX204), the company's most advanced candidate for type 2 diabetes, is in Phase 2 clinical trials. PPM-204 is a PPAR pan-agonist, targeting all three PPAR receptors, alpha, gamma and delta. In contrast to currently marketed PPAR compounds, PPM-204's activity on all three receptors is expected to modulate lipids and cholesterol, as well as lower glucose. The multi-targeted compound, designed to be less potent on each individual receptor and a partial agonist, should have broader therapeutic activity and a wider safety window than current therapies.
Since October 2004, Plexxikon has been co-developing PPM-204 with Wyeth. The companies are collaborating to identify additional development candidates for obesity.
Plexxikon is able to design PPAR compounds with activity against any of the three PPAR receptors or any combination of receptors, as well as vary the degree of agonism. Given this capability, the company is independently pursuing PPAR compounds with distinct profiles for the treatment of multiple sclerosis and inflammatory bowel disease.
