Plexxikon's Scaffold-Based Drug Discovery™ Platform To Integrate Synthematix' ARTHUR™ Technology

Research Triangle Park, NC & Berkeley, CA (January 15, 2003) – Synthematix, Inc., a software and informatics provider for synthetic chemists, and Plexxikon Inc., a structure-based drug discovery company, today announced that they have signed an agreement in which Plexxikon will incorporate Synthematix' proprietary ARTHURT technology as part of its drug discovery platform.

"Plexxikon's Scaffold-Based Drug DiscoveryT platform enables the discovery of novel scaffolds that can be rapidly synthesized into drug leads for new targets, addressing the key bottleneck in drug discovery today with novel and efficient chemistry. Synthematix' unique ARTHURT technology will provide us with the ability to build, search and mine synthetic chemical reactions and procedures to enhance and fully utilize our synthetic chemistry capabilities quickly and efficiently," said Dr. Michael Milburn, senior vice president of research for Plexxikon. "This technology also provides our chemists with a comprehensive solution for efficiently organizing, maintaining and enabling chemistry on our unique scaffolds."

The agreement provides Plexxikon with access to Synthematix' ARTHURT family of products consisting of two major components: a chemist-friendly database of synthetic organic reactions and experimental conditions combined with a unique search engine, and a suite of modules to enter, analog, annotate and mine the database. Together, the ARTHURT products allow chemistry organizations to create and electronically mine their own databases of chemistry with proven experimental conditions for the first time.

"We are pleased to be collaborating with Plexxikon, a leader in structure-based discovery of novel therapeutics," said Robin Y. Smith, chief technical officer and co-founder of Synthematix. "Plexxikon's innovative discovery approach is based on utilizing information derived from multiple technologies and across multiple disciplines. Synthematix' technology, built on an information-rich ReactionXMLTM format, allows data sharing, data mining and data visualization, essential components of a modern drug discovery effort."

Plexxikon Profile
Plexxikon is a leader in the discovery and development of novel small molecule pharmaceuticals to treat human disease. Plexxikon's proprietary Scaffold-Based Drug Discovery™ platform is being applied to build therapeutic franchises in metabolic and cardiovascular disease, inflammation and oncology. This discovery process integrates a number of state-of-the-art technologies, including structural screening as one key component that provides a significant competitive advantage over other drug discovery approaches. To date, the company has discovered a portfolio of clinical and preclinical stage compounds in each of these franchises.

Currently, the company's most advanced program is a PPAR pan-agonist for the treatment of diabetes and related cardiovascular complications, with the lead candidate now in Phase 2 clinical testing in collaboration with Wyeth Pharmaceuticals. Other discovery programs include a dual kinase inhibitor for the treatment of rheumatoid arthritis and other inflammatory diseases and a renin inhibitor for hypertension in collaboration with Servier. Plexxikon is seeking pharmaceutical and biotechnology partners for select collaboration opportunities. For more information, please visit www.plexxikon.com.
